I sold an item that was for collection only - the buyer then sent me a message saying he thought it was delivery so he wanted to cancel - didn’t think nothing off pressed cancel and thought all was well - then I realised all my money I have in my eBay fund was gone - the time was £600 but I had around £350 in my eBay account. What do I do!?

Assuming the buyer had paid, you have now refunded that payment with the cancellation.
Ebay will release the buyer's original payment to you. You have to wait for that.

I'm not sure if he did or not? All I know is he shortly after sent me a message asking to cancel I never received his funds but after cancelling it seems to have taken what was in my from other orders I had been saving.

The buyer must have paid and that's how ebay refund - from your funds and you keep the payment when it clears.

Sorry it’s a little confusing.
i sold an item for collection - £600
Within minutes he requested to cancel
i never received the £600 - it’s pending still
i had £375 in my eBay balance
its taken my £375 as a refund?! This had nothing to do with that sale it was in my account from other sales.
why has it taken this money? If he purchased then cancelled it surely it would have just given his pending payment back? Why has it taken my actual funds I don’t understand?
