If I have a variation listing for an item I have 200 in stock of and I sell it in quantities of 10, 20 and 50, does eBay keep track of the total stock?
I worry that someone could buy 4 lots of 50 and 2 lots of 20, and that would be more stock than I have.

No, ebay will reduce the listing quantities as they sell but they cannot calculate sales across different items listed even though the item is the same the packs are different so they are different items
ie you advertise qty 3 x 50 pack and sell 2 the quantity reduces to 1 x 50 pack but all the other pack sizes remain the same as advertised so qty 10 x 10 pack remains the same
Beware that when it relists the quantities can revert back to the original listed quantities because the listing is renewed back to what it was when you started
Simply manually change the quantities when they sell or only advertise the total in stock items split between the pack sizes

When we started selling multiple packs we got caught out with not keeping an eye on the multi listings and the relisting replenishing the stock so we only advertise the exact stock split across the packs and even then we sometimes get it wrong usually when stock runs low - when you have 5000 items it is less of a worry but when you get down to 100 items it's easy to slip up.
It depends on what level you restock at whether it needs more attention more often !
There are some programmes available to automatically calculate the stock levels across different platforms - we never bother the cost doesn't warrant it but you can look into it and make your own decision

Yes I think for now I'll just stick with the pack quantities I have up and offer a decent multi-buy for people who want bigger quantities.
Though currently eBay is so quiet (well for me anyway) I'm fairly new and still learning but frustrating when I know my stuff is extremely competitively priced but not getting the views or sales I expected.

It can take time to gain traction on ebay - a couple of pointers that might help or might not but they work for us.
Your photos are pretty good - but check them against google and bing photo requirements as ebay collaborate with google and they have to comply to be seen
Use the full space for the titles - use key words that you are aiming for - research the keywords - you are aiming for position 1 or 2 in ebay search - be aware that some of the top search words are difficult to gain position with as the big boys target them with huge resources - so maybe to start look at some of the less popular words to gain 1 or 2 spot with these - you can research the popularity and look at the phrases your buyers used to find you.
Maybe make it obvious the pack sizes / discounts
Make sure your item specifics are fully used and filled in
Keep your descriptions short - maybe a small bullet point list with the key words - test to see what your adverts look like on a mobile device / mobile app - optimise to make sure they view well and are easy to follow without flicking through page after page
Lastly sales means position - so sometimes take one or two items and price to sell - inch the price up as momentum grows
It is a lot of work to start with and it is constant to keep your listings in top spots - when you become popular - sellers will copy you and undercut you so you are constantly tweaking - but if you get visabilty right and hold your position you will sell at better prices than the rest at a higher volume as strange as that sounds !
