eBay have been running a zero final value fee (FVF) and zero insertion fee (IF) promotion since March 2020. This is a direct lift from the T&C's page:
1. Promotion Period
The zero Final Value Fees (“FVF”) and Insertion Fees (“IF”) on 250 listings per month promotion (“Promotion”) will be valid for new Business Seller accounts that register on www.eBay.co.uk(the “Site”) after 00:00:01 on 26th March 2020. The Promotion will be valid for (i) 90 days from the date you registered on the Site; or (ii) until 23:59:59 on 31st July 2020, whichever is earlier (“Promotion Period”) and is only valid for listings created on the Site. All times are as per the local UK time.
I'm a bit puzzled then as to why I owe eBay £400 fees! Am I missing something?
Brand new seller account and shop subscription (did change an existing personal account to a sellers one). Didn't start selling to take advantage of this promotion but seeing as it's there why not?!
Can't get in contact with eBay...any ideas?!

So you were already an Ebay seller, and simply upgraded your private account to business to take advantage of this promotion...? As the T&Cs clearly say for 'new business sellers', which I read as actual new business accounts, I imagine that's the reason you've been charged.
But I don't know for sure, so I suggest you post on the Business Seller board in the Discussion forums to see if anyone over there does. If not, you will have to contact Ebay (Help & Contact is at top and foot of any page). You can't currently phone them (Covid staff shortages), but Live Chat and Request-a-Callback are both available during working hours.
That you opened a shop too is irrelevant. Anyone can open a shop as lots of folk are listing stuff amassed over a lifetime of collecting. I personally think this option should not be available to private sellers as it's seriously abused, but that's another story!
