I've just had the buyer cancel the purchase of my item. It was at the stage were we had agreed on the price, the postage, I believe, had been paid and I'm assuming the money for the item (which hadn't been sent) would've been paid and was being held by ebay. What is the next step, and do I need to relist the item as it is showing up now as not active. Thanks.

Click on the dropdown menu arrow alongside the item on your orders page and select Cancel order > Buyer asked to cancel. The refund will be issued on the process. After that you can relist if you wish.

I don't think I've ever asked to cancel a purchase, but out of interest, if a buyer has paid for the item, then asks to cancel, how quickly does eBay refund?
Would it be quicker if the item happened to be paid for out of eBay balance funds...?

You mean from a a buyer point of view? If so, once the seller issues the cancellation, a refund is also issued straight away. Then you can expect your funds to be back in your original payment method in 1-2 business days, approximately.
