Hi, I sold my car on Sunday21st April, we arranged for the buyer to pick it up between 5-7 on Friday, but they failed to show; eventually that evening they contacted us and agreed to pick it up 9am but failed to show. Then, at 3am this morning they tried to call us twice 😬 what would you do in this circumstance?

Personally I would take a very dimview of any 3am phone calls.
It's your call, but maybe tell them you need the car collectiong by ***** date, and if they cannot manage that then you will cancel.
Then add them to your blocked list.

Have you contacted/phoned them back.
If the buyer hasn't paid yet,cancel using the buyer hasn't paid option,and you will get a final value fee credit and the buyer gets a non payment strike placed on there account,then block them here,https://www.ebay.co.uk/bmgt/BuyerBlock?

As already said, as buyer should not have paid at least until they have inspected the car, then treat as a non payer, and add buyer to blocked bidders list.
Sounds as though they have had a change of mind and are just wasting your time and the cost of the listing fee which is not refundable.
