Do you have a message from him confirming this?

What does the message say, can you post the main body of it here, leaving out any personal info?
Has he opened a return yet?

Does the care label say that it can be washed on a wool cycle?
If it does, and it's not handwash only, then you will have to accept the return.

You didn't help things by describing it as a knitted wool jumper, but stating that the material is 100% Mercerised Cotton. Mercerised Cotton can be machine washed so it's understandable that your buyer put it in a washing machine.
If the washing instructions say that it should be hand washed only it's a pity that you didn't include a photo, or at least state this in the description.
It's up to your buyer now to open a case under eBay's money back guarantee. How eBay will decide the case is up to them.
