Yes, you do. The charge is 4% of your reserve price, and has to be paid whether or not the item sells.
Using a reserve price is very expensive, especially if it doesn't sell and you have to relist it. Buyers don't like reserve prices either.
It's better and cheaper just to start the auction at the lowest price you would be willing to accept.
