As above seller want's to cancel an order he has already made. This is the second time this has happened. (not from the same buyer). I haven't sent it yet but I am tempted to tell him I have. The monitor is brand new there wouldn't be a genuine reason for him to return it.
I am tempted to tell him it has already been shipped and if he doesn't want it he will need to sell it on ebay.
What would you recommend?

Some times these happen twice a day, some times once a quarter, part of online selling
Cancel, use the opportunity and let them know youve cancelled but kindly ask them to consider you for future purchases, occassionaly they come back but also puts the platform in good light.
Then forgert and move on till the next time waster does it again

You could and given you are a private seller you could state no returns.
But you would leave yourself open to 1) negative feedback and 2) the risk they open an item not as described return that would be at your cost anyway.

Worst. Idea. Ever.
If a buyer wants to return a purchase, they are practically guaranteed return rights through ebay's money back guarantee.
If you force an unwanted purchase, you will likely recieve a SNAD return. There is also the possibility of a disgruntled buyer purposely damaging the monitor to justify a return.
Do both of you a favour and cancel the sale, as requested.

Would just cancel it and relist it again.
Would ask why they have had a change of mind though if its happened twice with 2 different buyers.

I would agree with @fatbobfan for the most part. Just accept the cancellation and re-list it.
However, I wouldn't invite the buyer to return. Quite the opposite. I wouldn't actually tell him, but I would add him to my blocked bidder list straight away. If he's mucked you about once, he'll think he can do it again.
