Ebay will not cancel the order , but I am afraid you will get a late dispatch mark against your name . This normally stays on your record for twelve months . You can try and get this removed but that is very unlikely. After the buyer requested you not to dispatch you should have explained sellers have to dispatch within a set time which would have been decided by yourself .

EBay won't cancel as the item has been paid for.
You could work with the buyer and cancel the order, without marking it as sent, and relist - you won't get a defect if the order is cancelled. Easy enough to relist, send the buyer the item number and have her re-buy when ready for combined invoice.
This is a regular situation for me, I normally would simply mark the item as sent and hold it to go with the other items, asking the buyer to leave feedback stating it arrived on time.
Sometimes though I forget and get the dreaded 'item overdue' - in this case I don't mark it as despatched - ever!
I just let the buyer know that it is with the other items and manually give her tracking if there is any, and still ask that she marks feedback as item arrived on time.
The listing will sit in 'awaiting despatch' and simply drop off 30 days - exactly as an unpaid item drops off the 'unpaid' list after 30 days. A wee eBay anomaly. And, as I don't like to look at the listing for 30 days, once I know it has been delivered and having no issues, I move the listing to Archive and forget about it.

Yes, I had this same situation recently. Just mail your buyer and tell them that you are marking the item as `Dispatched`, but you are not actually sending it until they notify you that they are ready. If they do not come back to you, or reply in a reasonable time, then just cancel the order.
