My buyer has requested I send Royal Mail but I have evri set up as simple delivery

how do I then mark as despatched and upload tracking once I have posted the item?

If you haven't sent it yet it's best to cancel/refund the transaction with the buyer's agreement (via the dropdown actions menu arrow for the item on your orders list) and relist with RM postage for the buyer to buy and pay again. 

You can temporarily opt out of using Evri because the buyer can't see what they're choosing  - here:
