Hi a buyer has bought and paid for a £4.00 item with £2.69 postage cost I was selling. They have selected the Yodel Store to Door postage option which is £6.19. I have tried contacting the buyer so that they can amend the postage option however they are not responding albeit I send a chaser message tonight. I'm concerned if I don't post the item tomorrow (post by date) that will negatively reflect on my rating. Can I cancel the order?

Yes, you can issue a cancellation via the dropdown menu for the item on your orders page. Use the reason "Something wrong with buyer's address", nearest we have to a tricky situation like this. The refund will be issued in the process.
And put their ID on your blocked bidder list:

You can opt out of Simple Delivery during the listing process if you're worried about this happening again. Use the Custom Postage or postage options icon on the right of the postage section.

Finally received a response from the buyer at 3am today which I picked up at 7am advising they can't amend the purchase. Advised them they can either cancel the order or I'll cancel. Will give them until later this morning before I head out & if I haven't received a cancellation from them then I'll cancel & block.

Buyer cannot cancel, you have to do it. Buyer can only ask you to cancel.
It is possible that your buyer lives in a remote area, where Yodel standard prices do not apply.
This is one problem when dealing with Couriers, only Royal Mail have same price for all parts of the UK.

Had another response from buyer basically suggesting I simply go to the Post Office rather than use the eBay delivery options. Have now cancelled the order and blocked the buyer. Something doesn't stack up.

The difference in postage is usually due to the buyer living in a 'remote zone' Scottish Highlands and Islands etc, which is why Yodel charge £6.19
