Sellers are supposed to post out according to there estimted delivery times so if you go beyond that time frame and a buyer marks the item as arriving later then yes you would get a late defect placed on your account.
Maybe look to cancel the transaction stating problem with buyers address,and list the item again for the buyer to buy, when they return from holiday

If you don't buy an eBay postage label or upload tracking and your buyer doesn't mark it as arriving later than the estimated delivery date when they come to leave feedback you won't receive a defect. You will be stuck with a 'dispatch your item' message for a while though (which you can safely ignore).
It's a bit cheeky of your buyer to ask this after they've bought the item though.
