When you make an offer should the seller respond to that offer ?

It is up to the seller and depending on the offer they may chose to ignore it.

£5 item offer of £1, cheeky offer and might be ignored, £5 item and offer of £6, then you will get a reply.

If no reply then assume that it is declined, no one can make a seller reply.

For politeness they should even if it's to decline or counter offer,  but some do not,  and it is certainly not mandatory to do so.

Offers are valid for 48 hours.  Do be careful not to bid or buy another in the meantime,  as you may end up with two to pay for.
