Recently sold an item and delivered less quantity than I should have (not 100% my mistake but it looked that like it was).
The buyer flamed me without giving me the chance to put things right.
Once I understood the issue the buyer received a full refund and got to keep the item.
I wonder if I should have asked for the feedback to be removed?

The only way to remove that feedback is to send the buyer a polite message apologising for the mistake and as it has been quickly remedied with a full refund, no need to send the item back, would they kindly complete a feedback revision you can send to them.

just put on a response as to what you did to rectify the situation. As a buyer i look at the negs and then at the response by the seller, they tell an awful lot about both parties.

I'm sure you have done so already, but, if not, be sure to add that buyer to your blocked bidders list.
No matter who's fault it was, you don't want to deal with this buyer again.

I was always under the impression that you had to ask, however, we got our very first non-positive in 20 years on a business account with 5000 feedback.
It was a fair comment - 'never arrived'. And it was a neutral, not a neg, so not even worth worrying about. Though at the time there has been no communication and no case opened. The case was opened about a week later and we refunded immediately.
A few days later the item arrived back with us, an untracked international sale, with 'unable to deliver' as the reason. So I went to feedback to inform the buyer and ask for a revision - nothing there.
So I wonder if, had you had asked the buyer to open a case and refunded through the case, the feedback would have been removed automatically? And I'm also wondering it only applied because it was a business account.
Not at all helpful here, of course, but might help in future?

Many thanks I had not thought of asking them to open a case and progress the refund from there (I just did it)...not sure if that would remove the negative feedback but it would not do my case any harm. The buyer was new to eBay (2 points feedback) so I put it down to inexperience!
Don't know how you can go 20 years without any negative feedback that's amazing.
If there is a next time I will progress them as you suggest thanks :>)
