Hi, I would like to purchase an item from the US. When I go to check out the following message is displayed “unfortunately, this seller won’t send this item to Derbyshire, United Kingdom”
because of this I am unable to purchase the item.
Any ideas why?

Sellers can select to sell internationally or not.
Have you messaged this seller? They may make an exception, put you on their buyer's exemption list and sell this item to you, but they are not obliged to.

That's all you can do , are there no UK sellers selling this item?
You can narrow the search to UK using that filter on the left sidebar when searching the item.

Any ideas why?
There are very good reasons why sellers may opt not to deliver to other countries.
One is that if a buyer "discovers" some reason why an item it is not as described, eBay will require the seller to refund the buyer's whole payment, including the cost of international delivery - and to pay for the return international delivery.
As eBay doesn't provide international prepaid return labels, the seller has to somehow arrange and pay for the item to be returned. The difficulty and cost involved, especially when the buyer refuses to assist, has led some sellers to give up and let the buyer keep the item for nothing - which was probably what they hoped for.
Personally, I'm amazed that so many sellers still deliver abroad. Perhaps they just don't realise the risks. But that's true of so many eBay users, both buyers and sellers.
